Epitalon vs DSIP: sleep and circadian research compared
Epitalon and DSIP both turn up in sleep and circadian-rhythm research, but they come at the topic from different directions. Epitalon (Ala-Glu-Asp-Gly) is a synthetic four-residue peptide modeled on a pineal gland extract, and its research centers on melatonin production and aging. DSIP, delta sleep-inducing peptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu), is a nine-residue peptide isolated from rabbits in the 1970s and studied for direct effects on deep, slow-wave sleep. Neither is an approved medicine, the evidence for both is old, small or concentrated in a few labs, and their US regulatory paths split in 2026: an FDA advisory committee backed Epitalon for pharmacy compounding but voted against DSIP.
- Structure: Epitalon is a tetrapeptide of about 390 g/mol; DSIP is a nonapeptide of about 849 g/mol.
- Research angle: Epitalon is studied for effects on pineal melatonin synthesis and circadian markers; DSIP for effects on slow-wave (delta) sleep itself.
- Evidence: mostly animal and cell work for both. Epitalon research comes largely from one St. Petersburg institute; DSIP's sleep role has been disputed since the 1980s.
- Mechanism: no receptor has been confirmed for either peptide.
- US status (October 2026): Epitalon is in the HHS 503A enforcement-discretion policy; DSIP (emideltide) is not, after a 6–7 committee vote.

Epitalon grew out of Soviet-era work on Epithalamin, a pineal gland extract that appears in the accessible literature from 1973. A 2025 review in the International Journal of Molecular Sciences (Araj and colleagues) summarizes the sleep-relevant findings:
- Rat pineal cells. In cultured rat pinealocytes, Epitalon affected AANAT (a key enzyme in melatonin synthesis) and the signaling protein pCREB, which suggested a direct effect on melatonin production. However, a separate study by Djeridane and colleagues found no effect on melatonin release from perifused rat pineal glands, so the cell data are not consistent.
- Aged monkeys. In old rhesus monkeys, Epitalon was reported to stimulate melatonin synthesis and normalize cortisol levels, two hormones with strong daily rhythms.
- A human biomarker study. In one placebo-controlled study of 75 women, urinary 6-sulfatoxymelatonin (a melatonin breakdown product) rose about 1.6-fold relative to placebo, and expression of several clock genes shifted. This measured biomarkers, not sleep quality.
- Telomeres and aging. Much of the Epitalon literature is about telomerase and cell aging, for example human fetal fibroblasts that kept dividing past passage 44 when controls stopped at 34. This is separate from sleep but explains why the peptide is often filed under longevity research.
The review itself flags the gaps: the mechanism is unclear, structural studies are very limited, and "information regarding critical issues about this peptide's safety is missing." Most human studies come from the St. Petersburg Institute of Bioregulation and Gerontology and are small.
DSIP: the slow-wave sleep angle
DSIP was reported by the Swiss group of Monnier and Schoenenberger, with key characterization papers in 1977. Unlike Epitalon, it was proposed as a sleep factor in its own right. A 1984 review in Neuroscience and Biobehavioral Reviews summarized the early animal data:
- In rabbits, rats, mice and humans it mainly promoted delta (slow-wave) sleep; in cats its effect on REM sleep was stronger.
- Its effects followed a U-shaped curve, so more was not more.
- DSIP-like material was detected in rat brain, peripheral tissues and the plasma of several mammals.
- Reported actions reached beyond sleep, to circadian and locomotor rhythms, hormone levels and neurotransmitters.
Later work was less tidy. In a 2001 Protein Spotlight profile, sleep researcher Alexander Borbély called DSIP a flop as a sleep substance, citing negative or ambiguous results, while co-discoverer Guido Schoenenberger argued the tests were conclusive and that funding, not data, ended the work. No DSIP gene has been found in rabbits and no receptor or precursor has been identified. Its half-life in vitro is about 15 minutes because an aminopeptidase-like enzyme breaks it down. A 2006 Journal of Neurochemistry review by Kovalzon and Strekalova summed up the field in its title: "a still unresolved riddle." Human reports exist, but they are older, small and largely uncontrolled.

Regulatory status in October 2026
Both peptides appear on FDA's bulk drug substance safety page among nominations later withdrawn, with the safety concerns shown above. In July 2026 the Pharmacy Compounding Advisory Committee recommended Epitalon for the 503A bulks list, while emideltide (DSIP) failed 6–7 with one abstention. On 28 October 2026 HHS announced a temporary enforcement-discretion policy for six peptides: BPC-157, Epitalon, KPV, MOTS-c, Semax and TB-500. DSIP is not among them. The policy is not an approval and does not add any peptide to the bulks list. See our FDA peptide vote explainer, the HHS compounding policy guide and are peptides legal? for detail.
Lab bench notes
For in-vitro stock solutions, a 1 mM solution needs about 0.390 mg of Epitalon or about 0.849 mg of DSIP per mL, using free-peptide molar masses. Salt forms weigh more, so use the formula weight on the batch certificate. The peptide calculator does the math, and how to verify a peptide CoA covers identity and purity checks. Frequently asked questions
What is the difference between Epitalon and DSIP?
Epitalon is a synthetic four-amino-acid peptide based on a pineal extract and studied for effects on melatonin synthesis and aging markers. DSIP is a nine-amino-acid peptide isolated from rabbits and studied for direct effects on slow-wave sleep. They share no sequence.
Which has more sleep research, Epitalon or DSIP?
DSIP has the more direct sleep literature, with EEG studies in several animal species, but its role has been disputed and little progress was made after the early 1980s. Epitalon research focuses on melatonin and circadian markers rather than measured sleep.
Are Epitalon and DSIP the same as melatonin?
No. Both are peptides, while melatonin is a small non-peptide hormone made in the pineal gland. Some Epitalon studies measured melatonin output as an outcome.
Are Epitalon or DSIP approved by FDA?
No. Neither is an approved drug. Epitalon is covered by a temporary HHS enforcement-discretion policy for 503A compounding announced on 28 October 2026; DSIP, listed as emideltide, was not recommended by the FDA advisory committee and is not covered.
Is there a known receptor for DSIP or Epitalon?
No receptor has been confirmed for either. For DSIP, no gene or precursor has been found in rabbits; for Epitalon, reviewers describe the mechanism as unclear.
