GLP-1 receptor agonists explained for researchers

A GLP-1 receptor agonist is a molecule that binds to and activates the glucagon-like peptide-1 receptor, a class B G protein-coupled receptor that native GLP-1 uses to signal. The receptor's mechanism links to glucose-dependent insulin secretion, lower glucagon secretion when glucose is high, slower gastric emptying, and satiety signals in the brain. Native GLP-1 lasts only minutes before the enzyme DPP-4 breaks it down, so drug molecules such as semaglutide are engineered to resist that enzyme and stay in circulation for days. This guide explains the incretin system, how GLP-1 compares with its partner hormone GIP, and how the licensed medicines differ from research-grade powders.

In short
  • Incretins are gut hormones released after eating that boost insulin release. The two main ones are GLP-1 and GIP.
  • Receptor: GLP-1R and GIPR are both class B, seven-transmembrane G protein-coupled receptors that signal through the adenylate cyclase/cAMP pathway.
  • The problem with the native hormone: DPP-4 degrades endogenous GLP-1 and GIP, and their half-lives are measured in minutes.
  • Engineered agonists: semaglutide resists DPP-4 and binds albumin, giving a half-life of about one week in its EU product information.
  • Licensed medicines are Ozempic and Wegovy (semaglutide) and Mounjaro and Zepbound (tirzepatide). Research-grade powder is not the approved product.

The incretin system in brief

The incretin concept dates to Creutzfeldt in the 1970s, who noted that glucose taken by mouth triggers a much larger insulin response than the same glucose given intravenously. The difference comes from hormones released by the gut in response to nutrients. Two hormones carry most of this effect:

  • GIP (glucose-dependent insulinotropic polypeptide), made by enteroendocrine K cells in the upper small intestine, the duodenum and jejunum.
  • GLP-1 (glucagon-like peptide-1), made by L cells spread through the lining of the distal gut, the ileum and colon.

A 2024 review in Frontiers in Endocrinology notes that both hormones are broken down by dipeptidyl peptidase 4 (DPP-4), that their half-lives are measured in minutes, and that only a small fraction of the active hormone reaches the general circulation. That short life is the central design problem for anyone trying to use the pathway pharmacologically.

How the GLP-1 receptor mechanism works

GLP-1R is a class B, seven-transmembrane G protein-coupled receptor. When an agonist binds, the receptor activates adenylate cyclase and raises intracellular cAMP, a pathway shared with the GIP receptor. The downstream effects described in the review literature and in regulator product information include:

EffectWhat is describedSource type
Insulin secretionStimulated in a glucose-dependent mannerEU product information (Ozempic)
Glucagon secretionLowered when blood glucose is highEU product information (Ozempic)
Gastric emptyingSlowed by GLP-1 infusion; a minor early post-meal delay with semaglutideReview; EU product information
AppetiteGLP-1 and GIP act on neurons in the brain's satiety centers; reduced appetite and energy intake with semaglutideReview; EU product information

The glucose dependence is an important point for experimental design. Because insulin release is stimulated mainly when glucose is raised, results in cell or islet models depend heavily on the glucose concentration used in the assay medium.

The blunted response to GIP in type 2 diabetes long made it the less obvious drug target. The development of dual agonists changed that picture. The same review describes tirzepatide as the only FDA-approved dual GIP/GLP-1 receptor agonist and notes it is designed to act more strongly at the GIP receptor than at the GLP-1 receptor.

How drug molecules outlast native GLP-1

Semaglutide is the clearest worked example of the engineering. The EU product information describes it as a GLP-1 analogue with 94% sequence homology to human GLP-1. RCSB PDB-101 lists three changes:

  • Alanine to Aib at position 8. Aib (α-aminoisobutyric acid) makes the peptide a much poorer DPP-4 substrate without significantly changing receptor binding.
  • Lysine to arginine at position 34.
  • Acylation of the lysine at position 26 with a spacer and a C-18 fatty diacid, which increases binding to albumin.

The EU product information gives a half-life of around one week and names albumin binding as the main mechanism of protraction. PDB-101 cites about 165 hours from the original discovery paper and roughly 13 hours for the earlier analog liraglutide. Our semaglutide vs tirzepatide vs retatrutide comparison sets the three molecules side by side.

Beyond GLP-1: dual and triple agonists

MoleculeReceptors targetedUS status (October 2026)
SemaglutideGLP-1Approved: Ozempic for type 2 diabetes (2017); Wegovy for chronic weight management (4 June 2021)
TirzepatideGIP and GLP-1Approved: Mounjaro for type 2 diabetes; Zepbound for chronic weight management (8 November 2023)
RetatrutideGIP, GLP-1 and glucagonInvestigational; not FDA-approved

Retatrutide adds the glucagon receptor as a third target. Its Phase 2 obesity trial was published in the New England Journal of Medicine in June 2023, and Phase 3 trials are covered in our TRIUMPH-4 explainer. The evidence for all three molecules comes from controlled clinical trials of the licensed or investigational products. That evidence does not transfer to unregulated powders of uncertain identity and purity.

Licensed medicines vs research-grade powder

The approved medicines are specific products: Ozempic and Wegovy (semaglutide) and Mounjaro and Zepbound (tirzepatide). Their manufacture, formulation and quality are reviewed by regulators. In the EU, for example, Ozempic's semaglutide is made in Saccharomyces cerevisiae cells by recombinant DNA technology.

Research-grade semaglutide, tirzepatide or retatrutide sold online is not the approved product. The FDA's page on unapproved GLP-1 drugs, updated 1 October 2026, says unapproved versions do not undergo FDA review for safety, effectiveness and quality. It names semaglutide, tirzepatide, retatrutide, survodutide and mazdutide sold "for research purposes" but marketed to consumers, and urges consumers not to buy them. It also states that retatrutide and cagrilintide cannot be used in compounding under federal law, and that salt forms such as semaglutide sodium and semaglutide acetate are different active ingredients from those in approved drugs. Frequently asked questions

What is the mechanism of a GLP-1 receptor agonist?

It binds and activates the GLP-1 receptor, a class B G protein-coupled receptor that raises cAMP. This is linked to glucose-dependent insulin secretion, lower glucagon when glucose is high, slower gastric emptying and satiety signaling in the brain.

What is the difference between GLP-1 and GIP?

Both are incretin hormones that act on class B GPCRs and are degraded by DPP-4. GLP-1 comes from L cells in the distal gut, GIP from K cells in the upper gut. GIP raises glucagon during hypoglycemia and its insulin effect is blunted in type 2 diabetes.

Why does native GLP-1 not work as a drug?

DPP-4 degrades it within minutes. Drug molecules such as semaglutide carry substitutions that resist DPP-4 and a fatty acid side chain that binds albumin, extending the half-life to about one week.

Is tirzepatide a GLP-1 receptor agonist?

It is a dual GIP and GLP-1 receptor agonist, designed to act more strongly at the GIP receptor. It is approved in the US as Mounjaro for type 2 diabetes and Zepbound for chronic weight management.

Is research-grade semaglutide the same as Ozempic or Wegovy?

No. Ozempic and Wegovy are regulator-approved products. The FDA says unapproved versions are not reviewed for safety, effectiveness or quality, and has warned about GLP-1 powders sold for research purposes.
